This species is common in the coastal area of Monterey Bay, California and is considered an important contributor to the seafood industry in that region. The cells are able to turn sunlight into chemical energy in the form of proton motive force by photosynthesis. The protons synthesized in the process of light-driven photosynthesis are thought to reduce ammonium to an ammonium ion by a reaction that is coupled to inorganic phosphate in the chloroplast. In our recent experiments, we found that the cells could maintain relatively high rates of growth at moderate levels of nitrate, but their growth was greatly inhibited when the concentration of nitrate was raised above 50 mM. The inhibition was found to be localized to the chloroplast, and increased by ammonium, indicating that inorganic nitrogen, in the form of nitrate, was actively reducing nitrate to ammonium. This process is different from the more well-known, internal N-uptake and assimilation pathway seen in higher plants, which relies on transmembrane transporters. The results were consistent with, and strongly supported, the hypothesis that the chloroplast plays a role in the assimilation of inorganic nitrogen. The data were further consistent with the idea that inorganic nitrogen is funneled down the metabolic pathways of the chloroplast to energy-synthesizing, photosynthetically active organelles, in P. umbilicalis. It is not clear why a higher organism such as Porphyra umbilicalis, which can be easily grown in a growth medium with low amounts of nitrogen, has evolved an internal pathway of nitrogen uptake, while most other microorganisms need a much higher concentration of nitrogen. Perhaps the nitrogen-use efficiency of our species was so high in the past that the internal pathway was not necessary.
